Michailova-McCulloch Model

Discussion

The Michailova-McCulloch model (Biophys J. 2001 Aug, 81(2):614-29) extends the Winslow Jaffri Rice model by adding the buffering by MgATP and MgADP as well as by calmodulin. The results show that
buffering has a modest effect on the Ca++ transient, lowering it slightly.

Comments:

  1. Michailova and McCulloch have noted that the model, both their version and that of Winslow et al 1999 and 2000, do not handle the Na balance and they have fixed Nai at 10 mM in the cytosol.
  2. The exchanges between the "subspace" (or the subsarcolemmal space between the apposed membranes of the terminal cisternae of the SR and the T-tubular sarcolemma) and the cytosol are calculated using time constants. A modified version translates these into conductances based on diffusion coefficients and distance in the cell, and have the advantage over the time constant in that the conductances can be set to zero to isolate the subspace events from cytosolic events. This will be made available as jbAP.mod.
